Gramicidin is a 15-residue, linear polypeptide antibiotic that forms conducting channels in lipid bilayers. These channels have been studied intensively for more than 25 years as model transport systems for large protein ion channels, high-resolution structural information on which is difficult to glean. The recent availability of high resolution structural data on gramicidin allows an interpretation of ion translocation through a channel at a molecular level. This book brings together an international and interdisciplinary group of experts whose work is focused on the gramicidin ion channel and on related ion channel peptides and proteins. It covers crystallographic, spectroscopic, electrophysiological and computational studies, and comparisons between insights derived from studies on gramicidin and on larger systems.
Correlations of Structure, Dynamics and Function in the Gramicidin Channel by Solid State NMR; X-ray Crystallographic Studies of Gramicidin; Design and Characterization of Gramicidin Channels with Side Chain or Backbone Mutations; Engineering Charge Selectivity in Alamethicin Channels; Non-contact Dipole Effects on Channel Permeation: Lorentzian Noise in Single Gramicidin A Channel Formamidinium Currents; Can We Use Rate Constants and State Models to Describe Ion Transport Through Gramicidin Channels?; The Equilibrium Properties of Ion Channels: Binding of Sodium to the Gramicidin Channel; Molecular Dynamics Simulations of Peptide Channels; Ionic Interactions in Multiply Occupied Channels; Peptide Influences on Lipids; Peptide-lipid Interactions and Mechanisms of Antimicrobial Peptides; Lessons From Porins; Regions Involved Ionic Selectivity of Nicotinic Acetylocholine and Related Receptors; The Gramicidin-based Biosensor - A Functioning Nano-machine.
