This clearly written and generously illustrated book provides fundamental material for advanced undergraduate and postgraduate studies on the molecular basis of enzyme catalysis. It is also a valuable resource for more advanced readers in biochemistry, chemistry, chemical biology, and molecular biology.
Transition States in Biological Chemistry provides an in-depth analysis of protein/transition state interactions and their role in catalysis for a mechanistically diverse set of enzyme-catalysed reactions. After describing modern experimental and computational methods for elucidating transition state structures, the core of the book presents case studies for specific enzymes. For each example, it explains the nature of the chemical challenge and its biological significance, followed by structural, biochemical and computational studies that provide insights into the stereoelectronic constraints and molecular strategies that underlie transition state stabilisation. It also explores how understanding transition state structure impacts on the discovery of drugs and herbicides, and the creation of designer enzymes exhibiting novel catalytic activities. Critical assessments of the state of current knowledge allow the identification and discussion of key opportunities in the field that will require further exploration.

Nigel Richards is Professor of Biological Chemistry at Cardiff University. He is well known for integrating computational and experimental methods to solve problems in mechanistic enzymology, and is a leading expert on oxalate and asparagine metabolism. His work has been described in over 130 papers and two US patents. Professor Richards is an elected Fellow of the American Association for the Advancement of Science (2010) and a Fellow of the Royal Society of Chemistry (2016). Mike Blackburn has been active in chemical biology research for over 50 years. He studied enzyme catalysis with W. P. Jencks and is an expert on the use of stable analogues of phosphate esters and anhydrides for studies on nucleotide-related metabolism and glycolysis. His most recent work has employed metal fluorides to generate transition state analogues for phosphoryl transfer reactions by kinases, mutases, and isomerases. He has authored some 300 research papers, several patents and one book (“Nucleic Acids in Chemistry and Biology”).
