It is now clear that in addition to its role within the cardiovascular system, the key mediator nitric oxide is also implicated in the normal physiological function and disease pathology of several organs and systems. This work summarizes the quantum leap from basic science to clinical applications emerging from a decade of research in the field.
Research on the key mediator nitric oxide has increased exponentially over the last ten years. It is now clear that, in addition to its role within the cardiovascular system, this mediator is also implicated in the normal physiological function and disease pathology of several organs and systems. A number of the fundamental research observations are now being developed into therapeutic principles and these are being pursued through clinical trials. This is the first work summarizing, in its two volumes, the quantum leap from basic science to clinical applications emerging from this decade of research.

Part A. Part 1 Generation and biological actions of nitric oxide: nitric oxide synthase gene regulation; localization and subcellular targeting of nitric oxide; molecular mechanisms of peroxynitrite reactivity; nitric oxide and gene transcription; regulation of nitric oxide synthase expression and activity by haemodynamic forces; mice deficient in eNOS and nNOS isoforms. Part 2 Nitric oxide and pathophysiology of diseases: hypertension; atherosclerosis - role of NO; nitric oxide and circulatory shock; kidney disease; the role of nitric oxide in preterm labour; multiple sclerosis; regulation of nitric oxide in human osteoarthritis-affected cartilage; the role of nitric oxide in rheumatoid arthritis; gastrointestinal function in shock. Part 3 Therapeutic applications: nitric oxide inhalation; nitric oxide synthase inhibitors; the pharmacology of peroxynitrite-dependent neurotoxicity blockade; gene therapy approaches with iNOS.
